QUESTIONS 1. (a)Explain the significance of ZnCl2 to acid phosphatase. ZnCl2 is dissociates into Zn2+ ions. Zn2+ ions are cofactors of acid phosphatase. The catalytic activity of the enzyme can enhances Zn2+ binding. Some enzymes are catalytically active without a requirement for additional ions or molecules. In others, activity depends on the presence of a non-protein cofactor. Cofactors fall into two major groups; they are either metal ions or organic compounds.

Acid phosphatase uses Zn2+ ions as cofactors. Tightly bound zinc atom contributes to the structural integrity of the polypeptide and a less tightly bound zinc atom that is required for catalysis. (b) Apart from ZnCl2, what other cation salt is important to acid phosphatase. Mg2+ ion is another ion that plays important role in acid phosphatase reaction. It works in a similar way with Zn2+ ions.

Mg2+ ions become the cofactors for acid phosphatase and stabilize the enzyme and maximize the activity. ZnCl2 saltcan be replaced MgCl2 salt. 2. In your opinion, will o-nitrophenol react with ALP under optimum conditions obtained? Explain. No, o-nitrophenol will not react with ALP under optimum conditions obtained. This is because ALP is a hydrolase enzyme responsible for catalyzing the cleavage of a phosphate group from a variety of compounds including nucleotides, proteins, and alkaloids.

Since o-nitrophenol doesn’t have phosphate group attached to it, so ALP will not react with it. CONCLUSION This chemical reaction is carried out by the enzyme ACP, Acid phosphate. Acid phosphatase is a phosphatase, a type of enzyme, used to free attached phosphate groups from other molecules during digestion. Many factors can influence the activity of acid phosphatase. The optimal reaction time is at minutes at which the reaction velocity is mol cm-3 / min.
